HRP in general is conjugated to IgG. In ELISA this IgG binds to specific antigen and then in presence of TMB and hydrogen peroxide, a soluble blue reaction product is produced. The reaction can be stopped using appropriate stop solution producing a soluble yellow reaction product. By measuring the concentration of the yellow product the amount of specific antigen can be determined.
Highlights
The HRP conjugated IgG can be kept stable in a proper storage buffer. We have successfully manufactured the storage buffer which contains either a protein, BSA or a non protein component. The buffer can be stored at 4- 8 degree Centigrade. Once a HRP Conjugate solution is prepared it should be kept at 4 – 8 degree centigrade. for at least one year.-
Instructions to use
The product can be used in ELISA. An appropriate conjugate solution is prepared in Medicore Diluent Stabilizer. About 50 to 100 microliters of diluted conjugate solution are added to each microwell and incubated. After incubation , the wells were washed and 50 - 100 microliters of Medicore TMB HRP Aqueous Substrate 1-component (Catalog # 1000, 1001 or 1002) added to each well. A soluble blue color reaction product is developed which can be stopped using an equal volume of 1N HCL, 0.18 m sulfuric acid or Medicore Stop solution (Cat# 2000). The blue color is changed to yellow color and can be read at 450 nm. The intensity of the color can be controlled using diluted antibodies or conjugates.
