TMB-HRPO ELISA Stop Solution is a propriety solution used to terminate the peroxidase/TMB reaction for ELISA applications. The TMB substrate reacts with immobilized horseradish peroxidase (HRP) conjugated secondary antibodies to produce a blue solution. Addition of the above STOP solution keeps the color range at 625- nm to 650 nm.

TMB Ready to use HRP Aqueous Substrate 1-Component contains 3 3’5 5’-tetramethylbenzidine in a mildly acidic buffer. The substrate is supplied as a Ready to Use solution containing hydrogen peroxide. When this substrate system is reacted with peroxidase, a soluble blue reaction product is produced. The reaction can be stopped using appropriate stop solution producing a soluble yellow reaction product, which is stable for at least one hour. By measuring the concentration of yellow product, the amount of reacting product can be detected.

The product can be used to block nonspecific binding sites to reduce background signals. After the coating step, the Synthetic Blocking Buffer is added to the microwells for blocking the remaining free binding sites. The blocking is complete and stable.
The blocking buffer eliminates the risk of false positive reactions in the assay and at the same time avoids the hazards related to the use of material like, bovine serum albumin. The Synthetic Blocking Buffer does not contain any proteins and thus cross reactivity can be avoided.

HRP in general is conjugated to IgG. In ELISA this IgG binds to specific antigen and then in presence of TMB and hydrogen peroxide, a soluble blue reaction product is produced. The reaction can be stopped using appropriate stop solution producing a soluble yellow reaction product. By measuring the concentration of the yellow product the amount of specific antigen can be determined.

The HRP conjugated IgG can be kept stable in a proper storage buffer. We have successfully manufactured the storage buffer which contains either a protein, BSA or a non protein component. The buffer can be stored at 4- 8 degree Centigrade. Once a HRP Conjugate solution is prepared it should be kept at 4 – 8 degree centigrade. for at least one year.-
Instructions to use
The product can be used in ELISA. An appropriate conjugate solution is prepared in Medicore Diluent Stabilizer. About 50 to 100 microliters of diluted conjugate solution are added to each microwell and incubated. After incubation , the wells were washed and 50 - 100 microliters of Medicore TMB HRP Aqueous Substrate 1-component (Catalog # 1000, 1001 or 1002) added to each well. A soluble blue color reaction product is developed which can be stopped using an equal volume of 1N HCL, 0.18 m sulfuric acid or Medicore Stop solution (Cat# 2000). The blue color is changed to yellow color and can be read at 450 nm. The intensity of the color can be controlled using diluted antibodies or conjugates.

We supply them as passage 1. These cells were washed, trypsinized and cryopreserved.
Propagation: DMEM (Mediatech, Inc) with 10% fetal bovine serum + 1% penicillin-Streptomycin (5,000 U/ml - 5,000 micrograms/ml).

Primary Mouse Embryonic Fibroblasts (PMEFs) are isolated from mouse embryos at E 12.5 day. These are primary cells and as such have a limited life span in culture. A healthy state of culture is maintained during cultivation. We used virus and pathogen free mice
Monolayers of mitotically inactivated feeder cells are essential for successful cultivation of mouse embryonic stem (ES) cells. ES cells are used in gene targeting and knock out mouse production. When grown under optimal conditions along with feeder cells, ES cells can be maintained in culture for many passages, retaining their non-differentiated morphology and pluripotent properties. The feeder cells secrete several important growth factors into medium that may play roles in maintaining the pluripotency of ES cells.
